Design and Synthesis of Pyridine-amide Based Compounds Appended Naproxen Moiety as Anti-Microbial and Anti-Inflammatory Agents

A series of pyridinamide-containing naproxen derivatives were synthesized through different routes starting with naproxenoyl chloride (2) which, upon reaction with 2-or 3-aminopyridine, gave the corresponding naproxenamide derivatives 3a,b. Also, urea derivatives 5a,b were obtained through the reaction of compound 2 with sodium azide followed by reaction with 2-or 3-aminopyridene via Curtius rearrangement. In addition, naproxenoyl isothiocyanate was synthesized and reacted with 2- or 3-aminopyridine, where the thiourea derivatives 7a,b were obtained. Furthermore, cyanoacetanilide 9 was synthesized and reacted with acetylacetone, arylidenemalononitriles or arylidenecyanoacetate to furnish the corresponding pyridine amide derivatives 10-13. All compounds were screened for anti-inflammatory activity using an in vivo rat paw edema model, from which six of them exhibited higher potency than naproxen. The ulcerogenic effect of most of the active compounds was also screened. In ulcerogenicity screening, the potent anti-inflammatory compounds 3b, 5a and 5b were devoid of any ulcerative effect. This is contrary to naproxen which caused 100% ulcerative effect on all tested animals. Structure-based molecular modeling described that the virtual screening agrees with the SAR of in vivo anti-inflammatory and ulcerogenic activities. Furthermore, all the synthesized compounds were screened for their anti-microbial activity.

Preliminary Studies on the Analgesic and Anti-Inflammatory Activity of Alcoholic Extract of Hibiscus Cannabinus Linn Seed

The analgesic and anti-inflammatory properties of Hibiscus cannabinus, a popular herb used for the management of pain and inflammation causing disorders was investigated in rats and mice. Significant antinociceptive was observed at higher dose of extract in writhing, tail immersion and hot plate animal models and anti-inflammatory activity was observed in carrageenan, serotonin and histamine induced paw edema in rats. The extract exhibit significant decreased paw edema in anti-inflammatory models. These results therefore indicate that Hibiscus cannabinus seed contains biologically active principles, which have potentials for the treatment of inflammatory processes.

Preliminary screening of anaphyllum wightii schott tubers for anti-inflammatory and antioxidant activity

To evaluate the preliminary phytochemical, antioxidant and anti-inflammatory activity of77 Anaphyllum wightii Schott. Bovine Serum Albumin denaturation inhibition assay was used for the evaluation of in-vitro anti-inflammatory activity and DPPH. Free radical scavenging and super oxide scavenging assays were used to assess the antioxidant activity of chloroform and aqueous extracts of the tubers of Anaphyllum wightii Schott. In anti-denaturation study, it was observed that chloroform extract showed greater percentage of inhibition of bovine serum albumin denaturation ie 49.2% whereas aqueous extract showed 38.4% at the 400µg/ml concentration respectively. In case of antioxidant screening also, chloroform extract showed better antioxidant power compared to aqueous extract dose dependently. Among the two extracts evaluated for anti-inflammatory and antioxidant activities chloroform extract of Anaphyllum wightii was found to possess significantly good anti-inflammatory and antioxidant activities and this can be attributed to the presence of alkaloids, saponins, flavonoids and phenolic compounds.

Phytochemical, Antioxidant and Antiinflammatory studies on the leaves of Putranjiva roxburghii

The In-vitro antioxidant and anti-inflammatory activity of the leaf extract of Putranjiva roxburghii were studied along with preliminary phytochemical investigation. The leaf of the plant contains mainly anthracene, cardiac and flavonoid glycosides along with phenolic compounds. Since many flavonoids have remarkable anti inflammatory activity the present work aims at evaluating the anti inflammatory activity of Putranjiva roxburghii by HRBC membrane stabilization. The ethanolic extract of the leaf was anlysed for antioxidant activity by DPPH method at different concentrations. Throughout the studies leaf extract showed potent antioxidant activity. Preliminary Studies on Anti-Inflammatory and Analgesic Activities of Jasminum Sambac (L.) Aiton in Experimental Animal Models

  Jasminum sambac (L.) Aiton (Oleaceae) is traditionally used as an antinociceptive and anti-inflammatory agent. The objective of this study was to investigate experimentally the possible analgesic and anti-inflammatory properties of Jasminum sambac. The effect of petroleum ether extract of leaves of Jasminum sambac (PEJS) was evaluated in experimental models of pain and inflammation. To Prepare a Poly Herbal Formulation Containing Pluchea lanceolata and Vitex negundo and Evaluate its Anti-Inflammatory Activity by Topical Application

  The aim of the study was to prepare a poly herbal formulation containing five herbs in the base of sesame oil and comparing its activity by using the carrageenan induced rat paw oedema model. The plants used in the study were Pluchea lanceolata, Vitex negundo, Solanum xanthocarpum, Cleorodandrum phlomoides and Curcuma longa. The oil obtained was applied topically to check the anti-inflammatory activity and comparing its efficacy with the standard which was marketed topical anti-inflammatory oil. The oil having more constituent of Vitex negundo in poly herbal formulation was found out to be having more potency in comparison to rest of the prepared samples as well as the marketed sample.
